It depends on your advisor or boss. It varies highly from one professor to another.

The amount of vacation and personal time is generous and available at the start. No build-up over several years!

The worst thing about this is that it is lab to lab and department specific. There isn't a university wide policy on this.

For full-time year-long academic staff, there is over 20 days of time off in addition to some paid holidays and flexible time off. Some departments are very small, so it can be difficult to take a bunch of time off in a row.

Excellent vacation and paid time off, markedly better than most other companies and organizations. This is a benefit that keeps employees at the UW-Madison, which makes them competitive with regard to other organizations/companies. However, as companies begin to have to make incentive packages for employees more attractive as appropriate candidates will begin to decline with baby boomers beginning to retire and job vacancies sprout up aplenty, this benefit in and of itself may not be enough to attract talent.
