#ImagingMassCytometry allows the detection of protein targets and provides general architectural details on #FFPE sections of both normal and cancerous tissues. The resolution of IMC permits visualization of proteins in the membranous, cytoplasmic and nuclear cell compartments. The benefit over traditional IHC techniques is the ability to #multiplex over 50 markers in a single experiment, enabling high-content analysis of human tissue with fewer tissue samples required. Scientists at Sapienza Università di Roma have made more headway in determining outcomes from #SARSCoV2 infection. The team looked at thyroid function, its association with SARS-CoV-2 severity, and clinical relevance of Low T3 syndrome. Using high-dimensional #singlecell analysis by #masscytometry, multiplex cytokine assay and #geneexpression profiling in PBMC, the team performed gene signature analysis and #immuneprofiling in #COVID19 patients. Thyroid function alterations appear to be frequently observed in the acute phase of COVID-19. Low FT3 serum values correlate with disease severity and an increase in the serum levels of markers for coagulation, inflammation and tissue damages, and so could be considered as a prognostic marker of COVID-19. https://lnkd.in/gbiZtYN

The tumor microenvironment plays a major role in high-grade serous ovarian cancer patient survival. To investigate how different cells interact to influence HGSC patients’ survival, a collaborative team developed a pipeline that combines #imagingmasscytometry, location-specific #transcriptomics, and deep learning to identify the distribution of various stromal, tumor and immune cells as well as their spatial relationship. #Breastcancer #genomics is known to play a role in drug response and resistance, but non-genomic tumor evolution can also generate phenotypic heterogeneity. Researchers at the Cancer Research UK (CRUK) used #masscytometry for high-throughput multi-parametric analysis of cellular heterogeneity and oncogenic signaling states in a biobank of patient-derived tumor xenograft models. The team developed a 33-marker #CyTOF panel, selected to profile markers relevant to breast cancer and to separate cells of murine and human origin. #Singlecell profiling revealed drug-induced cellular phenotypic dynamics and that pre-treatment cellular phenotypic composition is a determinant of anticancer therapy response. “Mass cytometry has provided unprecedented insights into normal tissue function and tumour biology, including the malignant cell-autonomous compartment and/or the tumour microenvironment,” the #publication states - https://lnkd.in/gvEWSuw
